Hypothesis
Can a combination of fed-batch and multiple induction strategies maximize GFP production in E. coli?
Conclusion
A pre-defined feeding and induction strategy increased the volumetric yield of GFP in E. coli without harming cell-specific production.
Supporting Evidence
- Cultures at 37°C produced GFP without needing to induce at 42°C.
- Multiple inductions increased GFP production significantly.
- A maximum specific GFP cell content of 60 mg GFP g-1 DW was achieved.
Takeaway
Scientists figured out how to make more of a special protein in bacteria by changing the temperature and feeding them at the right times.
Methodology
The study used a Monod kinetic model to analyze growth and GFP production in E. coli under different temperature and feeding conditions.
Limitations
The production of GFP was underestimated in simulations due to non-growth associated product formation under limiting nutrient conditions.
