Hypothesis

Hepcidin is expressed in airway epithelial cells and is regulated by early phase cytokines.

Conclusion

Airway epithelial cells express hepcidin in response to IFN-γ, but it does not significantly alter cellular iron transport.

Supporting Evidence

  • Hepcidin mRNA expression increased six-fold in response to pro-inflammatory cytokines.
  • IFN-γ induced hepcidin expression was sustained for 24 hours.
  • JAK2 phosphorylation was significantly activated by IFN-γ within 15 minutes.

Takeaway

Hepcidin is a protein that helps control iron in the body, and this study found that it is made by cells in the lungs when they are exposed to a certain signal, but it doesn't change how iron is moved around in those cells.

Methodology

Primary human bronchial epithelial cells were exposed to various cytokines and their hepcidin expression was measured using quantitative PCR.

Limitations

The study did not demonstrate a significant change in iron transport or ferroportin expression in response to hepcidin.

Participant Demographics

Healthy, nonsmoking volunteers aged 18-40 years provided alveolar macrophages.
