Hypothesis

Dicyanogold(I) may be useful for inhibiting HIV replication due to its ability to promote gold uptake by cells.

Conclusion

Dicyanogold(I) is readily taken up by H9 cells, leading to significantly higher intracellular gold levels compared to the external medium.

Supporting Evidence

  • External concentrations of 10 or 20 ppb had no effect on the growth rate of H9 cells.
  • Incubation in 50 ppb dicyanogold(I) resulted in an initial decrease in growth rate, but it returned to normal by day three.
  • Incubation with 100 ppb dicyanogold(I) resulted in a significant loss of cells, but the replication rate approached normal levels by day three.
  • The amount of gold taken up by H9 cells is at least 70 fold higher than the concentration in the external medium.
  • Preincubation with N-ethylmaleimide significantly inhibited the uptake of gold from dicyanogold(I).

Takeaway

The study shows that a gold compound can easily enter certain immune cells, which might help in fighting HIV.

Methodology

H9 cells were incubated with varying concentrations of dicyanogold(I) and analyzed using flow injection analysis with ICP-MS.
