Hypothesis

The study aims to examine the methylation status of the CpG sites in the cyclin D2 promoter region of gastric cancer and correlate it with cyclin D2 expression.

Conclusion

Promoter hypermethylation of cyclin D2 is associated with loss of cyclin D2 expression in gastric cancer.

Supporting Evidence

  • Hypermethylation at the CpG-rich region was detected in gastric cancer cell lines without cyclin D2 expression.
  • 65.2% of gastric cancers with cyclin D2 methylation had no cyclin D2 mRNA expression.
  • Only 12.5% of methylation-negative cases lost cyclin D2 mRNA expression.

Takeaway

This study found that when a certain gene (cyclin D2) is turned off in stomach cancer cells due to a chemical change (methylation), it can lead to cancer growth.

Methodology

The study used methylation-specific PCR and bisulphite DNA sequencing to analyze the methylation status of the cyclin D2 promoter in gastric cancer tissues and cell lines.

Limitations

The study does not explore other potential mechanisms for cyclin D2 loss in gastric cancer beyond methylation.

Participant Demographics

30 males and 17 females with a mean age of 64.8 years.
