Hypothesis
The study investigates the effect of co-solvents Cremophor EL and polysorbate 80 on the absorption of orally administered paclitaxel.
Conclusion
Cremophor EL significantly limits the absorption of orally administered paclitaxel compared to polysorbate 80.
Supporting Evidence
- Paclitaxel formulated in polysorbate 80 resulted in a significant increase in the maximal concentration and area under the concentration–time curve compared to Cremophor EL.
- When formulated in Cremophor EL, paclitaxel excretion in faeces was significantly higher than for the polysorbate 80 formulation.
Takeaway
This study found that a substance called Cremophor EL makes it harder for cancer patients to absorb a medicine called paclitaxel when taken by mouth.
Methodology
Six patients received oral paclitaxel dissolved in either polysorbate 80 or Cremophor EL in a randomized setting.
