Hypothesis
The study aims to characterize the core histone genes of Giardia intestinalis, including their genomic organization, promoter structure, and expression.
Conclusion
Giardia intestinalis has a single class of core histone genes that are constitutively expressed at relatively high levels, and their mRNAs are polyadenylated.
Supporting Evidence
- Two copies each of the core histone H2a, H2b, and H3 genes, and three copies of the H4 gene were identified.
- The transcription start sites for the histone genes were located near the start of their coding regions.
- The core histone genes are constitutively expressed at approximately equivalent levels.
Takeaway
This study found that Giardia has fewer types of histone genes than other organisms, and it doesn't seem to need a specific histone for DNA packing.
Methodology
The study involved identifying the copy number, chromosomal localization, and transcription initiation sites of the core histone genes, along with promoter analysis using luciferase reporter assays.
Limitations
The study could not locate a gene encoding the linker histone H1, raising questions about its necessity in Giardia.
