Hypothesis
To investigate the intra-species diversity of S. flexneri genomes further, we have sequenced the complete genome of S. flexneri 5b strain 8401 and compared it with S. flexneri 2a.
Conclusion
The two S. flexneri genomes are very similar, suggesting a high level of structural and functional conservation, with differences reflecting different selection pressures during evolution.
Supporting Evidence
- The Sf8401 chromosome is 4.5-Mb in size, smaller than Sf301 due to the absence of the SHI-1 pathogenicity island.
- There are 6 inversions and one translocation in Sf8401 compared to Sf301.
- The genomes share over 97% of their sequences, indicating high conservation.
Takeaway
Scientists looked at the DNA of two types of Shigella bacteria to see how they are alike and different, which helps us understand how they cause sickness.
Methodology
The complete genome sequence of S. flexneri 5b strain 8401 was obtained through shotgun sequencing and compared with S. flexneri 2a strain Sf301.
Limitations
The study primarily focuses on genomic comparisons and may not address all aspects of pathogenicity or environmental factors.
