TLR-ligands can make dendritic cells tolerant to TNF gene expression by blocking signal transduction through IRAK-1.

  • DCs stimulated with TLR4 ligand LPS showed rapid TNF expression.
  • Pre-culturing DCs with LPS reduced their response to subsequent stimulation.
  • IRAK-1 levels were significantly lower in LPS-tolerant DCs.

Takeaway

When dendritic cells are exposed to certain signals too many times, they stop responding as strongly. This study shows that two specific signals can make them less active.

Methodology

Dendritic cells were generated from PBMC and stimulated with TLR ligands to assess TNF expression and IRAK-1 levels.
