Hypothesis

LY 255283 may inhibit the production of leukotriene B and thromboxane B2 by antagonizing the effect of ionophore-induced leukotriene B on specific enzymes in human immune cells.

Conclusion

LY 255283 significantly inhibited the synthesis of leukotriene B4 and thromboxane B2 in human peripheral blood polymorphonuclear leukocytes and monocytes.

Supporting Evidence

  • LY 255283 inhibited the production of leukotriene B4 in human peripheral blood polymorphonuclear leukocytes.
  • LY 255283 also inhibited the production of thromboxane B2 in monocytes activated by calcium ionophore.
  • The study showed that the effect of LY 255283 was more prominent on the synthesis of leukotriene B4, especially in monocytes.

Takeaway

This study found that a drug called LY 255283 can stop certain cells in our blood from making substances that cause inflammation.

Methodology

Human peripheral blood polymorphonuclear leukocytes and monocytes were isolated from healthy volunteers and incubated with LY 255283 before stimulation with calcium ionophore A23187 to measure the production of leukotriene B4 and thromboxane B2.

Limitations

The effects of LY 255283 on 5-lipoxygenase and LTA hydrolase activities were not studied.

Participant Demographics

Healthy volunteers who had abstained from any drugs for at least 1 week.
