Hypothesis
Interactions between RHAU and different G4 dimers are worthy of extensive exploration.
Conclusion
RHAU has weak interactions with the intramolecular hybrid tandem unstacked dimer formed by D-24TTG, while the interaction with the Z-G4 dimer is much weaker due to steric hindrance.
Supporting Evidence
- RHAU exhibited stronger interactions with intermolecular stacked/interlocked parallel dimers than with intramolecular tandem stacked parallel dimers.
- The hybrid tandem unstacked G4 dimer could form special binding sites, leading to a weak interaction with RHAU.
- Steric hindrance at terminal G-tetrads greatly weakened interactions with RHAU.
Takeaway
This study looks at how a peptide called RHAU interacts with different structures of DNA called G-quadruplexes, finding that some shapes work better than others.
Methodology
Interactions were investigated using size-exclusion chromatography (SEC), circular dichroism (CD), ultraviolet–visible absorption spectroscopy (UV–Vis), and native polyacrylamide gel electrophoresis (native-PAGE).
Limitations
The study primarily focuses on specific G4 dimers and may not represent all possible interactions with RHAU.
