Hypothesis
The phenyl group Phe12 of deacetyl-thymosin b4 plays a crucial role in restoring the impaired blastogenic response of uraemic T-lymphocytes.
Conclusion
The analogue [1-Nal12]deacetyl-thymosin b4 showed a stronger restorative effect on impaired T-lymphocytes than deacetyl-thymosin b4, while [Cha12]deacetyl-thymosin b4 had no restorative effect.
Supporting Evidence
- The analogue [1-Nal12]deacetyl-thymosin b4 exhibited stronger restorative activity than deacetyl-thymosin b4.
- [Cha12]deacetyl-thymosin b4 showed no restorative effect on T-lymphocytes.
- Uraemic patients had significantly lower T-lymphocyte responses compared to healthy controls.
Takeaway
Researchers tested new versions of a protein to see if they could help sick immune cells work better, and one version worked better than the original, but another didn't help at all.
Methodology
Two thymosin b4 analogues were synthesized and their effects on the blastogenic response of T-lymphocytes from uraemic patients were evaluated using a fluorometric blast-formation test.
Limitations
The study involved a small sample size of only three uraemic patients.
Participant Demographics
Three uraemic patients needing dialysis treatment, suffering from recurrent infections.
