Hypothesis
The expression of PPARγ is underexpressed in PAX8/PPARγ-negative thyroid tumors.
Conclusion
The study found that PPARγ expression is usually underexpressed in multiple types of thyroid tumors, which may be important in the development of thyroid neoplasias.
Supporting Evidence
- PPARγ expression was significantly lower in follicular thyroid carcinomas compared to normal tissue.
- 25% of follicular thyroid carcinomas and 17% of follicular adenomas expressed the PAX8-PPARγ fusion gene.
- Most thyroid tissues expressed PPARγ1, PPARγ2, and PPARγ4.
Takeaway
This study looked at thyroid tumors and found that a protein called PPARγ is often not present in these tumors, which might help explain how they grow.
Methodology
The expression of PPARγ was analyzed using RT–PCR, interphase FISH, real-time RT–PCR, and immunohistochemistry.
Limitations
The study did not perform quantitative RT–PCR to distinguish wild-type transcripts from PAX8-PPARγ fusion mRNAs.
