Hypothesis

The spectrum of TP53 mutations in regions of low incidence of oesophageal squamous cell carcinoma is unknown.

Conclusion

SCCE from areas of high and low incidence present a similar pattern of molecular alterations but differ by the type of TP53 mutations.

Supporting Evidence

  • TP53 mutations were detected in 36% of the cases (12/33).
  • P63 was amplified in 5/32 cases tested (15.5%).
  • No amplification of MDM2 was found.
  • Frequent loss of p16INK4a/CDKN2a expression was observed in 46% of cases.
  • Frequent loss of p27Kipl expression was observed in 25% of cases.
  • Cyclin E was overexpressed in 70% of cases.
  • Cox2 was overexpressed in 42% of cases.

Takeaway

This study looked at cancer samples from a place where oesophageal cancer is rare and found that while the types of mutations are different, the overall patterns of cancer changes are similar to areas where the cancer is more common.

Methodology

TP53 mutation analysis, MDM2 and P63 amplification testing, and expression studies of various proteins.

Participant Demographics

Tumours collected in Lyon, an area of low incidence.
