Hypothesis
The inhibition of nitric oxide release by prostaglandin E2 and iloprost in lipopolysaccharide-activated J774 macrophages may be secondary to the inhibition of tumor necrosis factor generation, mediated by cAMP.
Conclusion
Prostaglandin E2 and iloprost inhibit nitric oxide and tumor necrosis factor release in macrophages through a mechanism involving cAMP.
Supporting Evidence
- PGE2 and iloprost inhibited nitric oxide release in a concentration-dependent manner.
- The inhibition of TNF release was also concentration-dependent.
- Dibutyryl cAMP mimicked the effects of prostaglandins.
- The selective phosphodiesterase inhibitor RO-201724 enhanced the inhibitory effects of both PGE2 and iloprost.
Takeaway
This study found that certain substances can help stop immune cells from making harmful chemicals that can damage tissues.
Methodology
J774 macrophages were activated with lipopolysaccharide and treated with various concentrations of prostaglandins and cAMP to measure nitric oxide and TNF release.

Murine monocyte macrophage cell line J774
